After the initial immunization, test-bleed mice are repeatedly injected with the antigen in different combinations and titrations to generate antibody titers suitable for fusion and selection. The resulting hybridoma cells are fused with selected positives in selective HA medium, and the resultant antibodies are screened via ELISA, flow cytometry, and immunohistochemistry to select high-quality, functional, and specific antibodies.
Once the antibodies have been selected, they are subjected to limiting-dilution subcloning to ensure monoclonality. The resulting cell lines are expanded and cultured to produce antibody-containing supernatants. These are then filtered, concentrated, and purified using Protein A or Protein G resin to produce a final monoclonal antibody product with a desired isotype. This is then subjected to extensive testing under various assay and storage conditions to ensure long-term reagent stability and fidelity.
